The study evaluated different vaccine formulations incorporating Leishmania amazonensis antigens (LaAg) combined with various adjuvants, including complete Freund’s adjuvant (CFA), saponin (SAP), and AddaVax™, to enhance immunogenicity and protective efficacy in BALB/c mice.  

Among the tested formulations, the combination of LaAg with SAP was the most effective. It significantly induced a delayed-type hypersensitivity (DTH) response, increased IFN-γ-producing CD4+ and CD8+ T cells in draining lymph nodes, and resulted in reduced lesion size and lower parasite burden compared to other formulations. 

The study concluded that saponin-based adjuvants significantly enhance the immunogenicity and efficacy of LaAg, converting it from a non-protective to a partially protective vaccine against L. amazonensis infection. The LaAg + SAP formulation could be a potential candidate for further development in leishmaniasis vaccine research.
